Theophylline and doxapram are used to treat apnea of prematurity, which occurs frequently to low birth weight infants, although theophylline has no indication and doxapram has a contraindication for the infants. To provide individualized optimal dosage regimens for these drugs, we are conducting clinical studies to elucidate population pharmacokinetics of the drugs in the infants, as a member of the Neonatal Pharmacology Research Network. Acute respiratory distress syndrome (ARDS) is a severe lung disease with pulmonary vascular hyperpermeability, and has a high mortality. However, there is little drug useful for ARDS. Therefore, we are trying to find candidates of drugs for the prevention and treatment of ARDS and to elucidate the pathogenesis of the lung injury, using oleic acid-induced lung injury, which is an animal model of the disease.
